A method for the simultaneous determination of gamma-aminobutyric acid, putrescine, spermine, spermidine and cadaverine is described. A precolumn derivatization procedure utilizing omicron-phthalaldehyde-beta-mercaptoethanol is employed to form electroactive isoindole derivatives, which are separated by reversed-phase gradient elution chromatography and detected amperometrically (+ 650 mV vs. Ag/AgCl) at a glassy carbon electrode. By careful optimization of the experimental conditions, the baseline drift during the gradient run could be contained within 0.5 nA. On-column detection limits are typically in the low pmol range and linear responses in the 5-200 pmol injected range. The procedure has been applied to rat brain, kidney and liver tissues after homogenization with perchloric acid.
