Signaltransducerandactivatoroftranscription3(STAT3)regulatesmanycriticalfunctionsinhumannormalandmalignanttissues,suchasdifferentiation,proliferation,survival,angiogenesisandimmunefunction.ConstitutiveactivationofSTAT3isimplicatedinawiderangeofhumancancers.Assuch,STAT3hasbeenstudiedasatumortherapeutictarget.ThelastapproachusingsmallmoleculeSTAT3inhibitorshasbeenthemostexaminedsoHeterocylesandCarbazole.Heterocylesareanessentialclassofmolecules,assumingaroleinmanyaspectofourlife.Indeedheterocyclicnucleusisacommonfeatureofseveralbiomoleculeandbioactivecompoundsincludingagrochemicalproductsanddrugs.Inthisreviewwefocusedontwoimportantclassofheterocycliccompounds:carbazolesandNHCs(N-heterocycliccarbenes)andthereimplicationsincancerinhibitions.Carbazoles,prevalentasstructuralmotifsinvarioussyntheticmaterialsandnaturallyoccurringalkaloids,asisknown,havemanyapplicationsespeciallyaspromisingbioactivecompoundsduetotheirbiologicalproperties,knownsince1965.FurthermoreNHCsareaclassofstablecarbenesthatoverthelastfewyearshaveenteredthefieldas“new”ligandsforbioactivecoordinationcompounds.IthasbeendemonstratedthatmetalNHCcomplexescanbeusedtodevelophighlyefficientmetalbaseddrugswithpossibleapplicationsinthetreatmentofcancerorinfectiousdiseases.
